LL-37 is the active 37-amino-acid form of the human cathelicidin antimicrobial peptide (hCAP18). It is a cationic, amphipathic α-helical peptide produced by neutrophils, epithelial cells, and other immune cells. It serves as a key component of the innate immune system, providing broad-spectrum defense against bacteria, viruses, fungi, and parasites while also modulating inflammation and promoting tissue repair.
Mechanism of action
LL-37 exerts its effects primarily through membrane disruption: its amphipathic structure allows it to insert into negatively charged microbial membranes, forming pores or causing carpet-like destabilization that leads to leakage and cell death. Beyond direct antimicrobial activity, it modulates immune responses by interacting with receptors (e.g., TLRs, FPRL1), inhibiting pro-inflammatory cytokine release in some contexts while promoting chemotaxis and angiogenesis. It also enhances wound healing by stimulating cell migration, proliferation, and extracellular matrix remodeling.
